Molecular Architecture of Early Dissemination and Massive Second Wave of the SARS-CoV-2 Virus in a Major Metropolitan Area

Abstract

We sequenced the genomes of 5,085 SARS-CoV-2 strains causing two COVID-19 disease waves in metropolitan Houston, Texas, an ethnically diverse region with seven million residents. The genomes were from viruses recovered in the earliest recognized phase of the pandemic in Houston, and an ongoing massive second wave of infections. The virus was originally introduced into Houston many times independently. Virtually all strains in the second wave have a Gly614 amino acid replacement in the spike protein, a polymorphism that has been linked to increased transmission and infectivity. Patients infected with the Gly614 variant strains had significantly higher virus loads in the nasopharynx on initial diagnosis. We found little evidence of a significant relationship between virus genotypes and altered virulence, stressing the linkage between disease severity, underlying medical conditions, and host genetics. Some regions of the spike protein - the primary target of global vaccine efforts - are replete with amino acid replacements, perhaps indicating the action of selection. We exploited the genomic data to generate defined single amino acid replacements in the receptor binding domain of spike protein that, importantly, produced decreased recognition by the neutralizing monoclonal antibody CR30022. Our study is the first analysis of the molecular architecture of SARS-CoV-2 in two infection waves in a major metropolitan region. The findings will help us to understand the origin, composition, and trajectory of future infection waves, and the potential effect of the host immune response and therapeutic maneuvers on SARS-CoV-2 evolution.

Coronavirus mutation emerges that may bypass mask-wearing, hand-washing protections

A new COVID-19 mutation appears to be even more contagious, according to a study — and experts say it could be a response by the virus to defeat masks and other social-distancing efforts.

Scientists in a paper published Wednesday identified a new strain of the virus, which accounted for 99.9 percent of cases during the second wave in the Houston, Texas, area, the Washington Post reported.

The paper, which has not been peer-reviewed, said people with the strain, known as the D614G mutation, had higher loads of virus — suggesting it is more contagious.

Virus delays Rio’s Carnival for first time in a century

Rio de Janeiro delayed its annual Carnival parade, saying Thursday night that the global spectacle cannot go ahead in February because of Brazil’s continued vulnerability to the pandemic.

Rio’s League of Samba Schools, LIESA, announced that the spread of the coronavirus has made it impossible to safely hold the traditional parades that are a cultural mainstay and, for many, a source of livelihood.

“Carnival is a party upon which many humble workers depend. The samba schools are community institutions, and the parades are just one detail of all that,” Luiz Antonio Simas, a historian who specializes in Rio’s Carnival, said in an interview. “An entire cultural and productive chain was disrupted by COVID.”

Rio’s City Hall has yet to announce a decision about the Carnival street parties that also take place across the city. But its tourism promotion agency said in a statement to The Associated Press on Sept. 17 that without a coronavirus vaccine, it is uncertain when large public events can resume.

Infectious coronavirus found on salmon for up to a week, study suggests

While the Centers for Disease Control and Prevention and the Food and Drug Administration have said there’s no evidence of contracting the coronavirus from food or its packaging, a new study suggests salmon could potentially carry traces of COVID-19.

A new study from Chinese researchers suggests that traces of COVID-19 on salmon were detected and could be infectious for more than a week.

Scientists from South China Agricultural University and Guangdong Academy of Agricultural Sciences in Guangzhou collected virus samples from salmon and noted it could survive up to eight days at 39 degrees Fahrenheit. The report, released last week, is still awaiting peer review and publication.

You are correct. COVID-19 is the disease caused by the virus SARS-CoV-2. Most news agencies and most people say COVID when they means SARS-2. The article does say the word "virus", so the mistake is probably editorial.

We wear face coverings, socially distance, and other things to reduce or MITIGATE my chances of getting COVID.

Severity of coronavirus infection may be determined by face mask use, study suggests

Facemasks may limit the severity of coronavirus cases, University of California San Francisco researchers said in a paper published in the Journal of General Internal Medicine.

Although face coverings are more effective in preventing droplets from being expelled into the air than they are in preventing droplets from being breathed in, masks still may be able to mitigate how severe an illness an individual gets if infected by COVID-19, researchers explained in the release.

Anything that reduces the viral load is good.

The table below shows confirmed cases of COVID-19 reinfection. The first confirmed case of reinfection was reported in Hong Kong on August 24, and six others were reported later that week. Earlier cases of suspected reinfection could not be confirmed due to a lack of data.

People with SARS-CoV-2, the virus which causes COVID-19, may continue to test positive for several months without being sick or infectious. A reinfection is confirmed when testing shows each virus’ genetic makeup is different to a degree which cannot be explained through in-vivo evolution.

Table at link.

16 reinfections. And having it once doesn't seem to prevent you from having a severe case the second time.

Efficacy and Safety of Hydroxychloroquine vs Placebo for Pre-exposure SARS-CoV-2 Prophylaxis Among Health Care Workers

A Randomized Clinical Trial

Finding

In this double-blind, placebo-controlled randomized clinical trial that included 132 participants and was terminated early, there was not a significant difference in reverse-transcriptase polymerase chain reaction–confirmed SARS-CoV-2 incidence between hydroxychloroquine and placebo cohorts.

Meaning

Among hospital-based health care workers, daily hydroxychloroquine did not prevent SARS-CoV-2 infection, although the trial was terminated early and may have been underpowered to detect a clinically important difference.

Face masks don't restrict oxygen or contribute to carbon dioxide buildup: study

Breathe easy. A new study reiterates that surgical face masks don’t cause a buildup of carbon dioxide or restrict oxygen, despite opposing claims.

[snip]

The small study included 15 military veterans with severe COPD, each with lung function under 50%, and 15 healthy participants. All participants wore masks for 30 minutes and were told to walk for six minutes while wearing the surgical masks. Researchers then gave each participant a blood test and discovered there were no differences in levels of oxygen or carbon dioxide.

“This data find that gas exchange is not significantly affected by the use of surgical mask, even in subjects with severe lung impairment,” Campos said in the study.

News Release 2-Oct-2020

Face masks unlikely to cause over-exposure to CO2, even in patients with lung disease

American Thoracic Society

New research findings contradict statements linking wearing face masks to carbon dioxide poisoning by trapping CO2. During the COVID-19 pandemic the wearing of face masks has become a highly political issue with some individuals falsely claiming that wearing face masks may be putting people's health at risk. The study published in the Annals of the American Thoracic Society shows otherwise.

Effect of Face Masks on Gas Exchange in Healthy Persons and Patients with COPD

It is important to inform the public that the discomfort associated with mask use should not lead to unsubstantiated safety concerns as this may attenuate the application of a practice proven to improve public health. As growing evidence indicates that asymptomatic individuals can fuel the spread of COVID-19,12 universal mask use needs to be vigorously enforced in community settings, particularly now that we are facing a pandemic with minimal proven therapeutic interventions. We believe our data will help mitigate fears about the health risks of surgical mask use and improve public confidence for more widespread acceptance and use.

Those with lung problem know better. Experience is better than research.
By the time they got around to taking the blood, they might not have seen the difference, but it exists.
And six minutes of exertion is not enough.

Try 30 or 60 min, then test.
No they won't do that, as they know the result and it won't fit their agenda!
